Effect of manganese on bovine sperm motility, viability, and lipid peroxidation in vitro

چکیده
Manganese (Mn) is a chain-breaking antioxidant in biological systems. The objective of the present study was to determine the optimal dose of Mn to reverse free radical-mediated oxidative damage on motility, viability, and lipid peroxidation (LPO) of sperm from five bulls (local crossbred cattle). Fresh semen was suspended in 2.9% sodium citrate, divided into equal fractions, and subjected to Mn treatment (0, 60, 100, or 200 μM) in the presence or absence of an oxidative stress inducer (ferrous ascorbate, FeAA; comprised of 150 μM FeSO4 and 750 μM ascorbic acid). All sperm suspensions were incubated (37°C) for 2 h. Treatment with FeAA decreased motility and viability but increased lipid peroxidation. All doses of Mn increased motility and viability but decreased LPO; however, 60 μM Mn was most effective. For sperm motility, viability, and LPO level, there were significant main effects of bull, treatment, and interval, as well as their interactions. In conclusion, Mn reduced the oxidative stress (LPO) caused by FeAA and improved sperm motility and viability under in vitro conditions as well as under induced oxidative stress.

P-85: Detrimental Effects of High Concentration Taurine on Sperm Motility and Viability
Background: Taurine regulates an unusual number of biological phenomena, including heart rhythm, contractile function, blood pressure, platelet aggregation, neuronal excitability and body temperature. It appears to be a sperm motility factor (SMF), although the mechanism that maintains motility has apparently not been elucidated. It may act by alterations in either ion transport (osmoregulation)...
متن کاملManganese provides antioxidant protection for sperm cryopreservation that may offer new consideration for clinical fertility
Reactive oxygen species (ROS) are generated by sperm metabolism. While, ROS are required for maturation, capacitation and acrosome reaction, they also modify many peroxidable cellular compounds. There is production of ROS during cryopreservation and frozen spermatozoa are highly sensitive to lipid peroxidation (LPO). Antioxidants exert a protective effect on the plasma membrane of frozen bovine...
متن کاملCadmium as an etiology of sperm dysfunction in Holstein bulls
Oxidative stress has been identified as a crucial factor leading to male infertility largely due to peroxidativedamage to the sperm cell membrane. Cadmium (Cd) is a widespread environmental pollutant and oneof the well-known reproductive toxicants. Because of its long biological half-life (10–30 years), Cdaccumulates in the biological systems. The present study was designed to assess the concen...
متن کاملSynergistic Protection of L-Arginine and Vitamin E On Lipid Peroxidation of Asthenospermic Patients
Background Lipid peroxidation is known to cause various impairments to sperm cells and may play a major role in the etiology of male infertility. Asthenospermia is the main factor of male infertility and has significantly higher level of peroxidation than in normozoospermic males. MaterialsAndMethods Using thiobarbituric acid (TBA) assay procedure, we have determined the level of lipid peroxida...
